Yes! Immensely. Most especially on long runs in 6" Pipe.
Even in 10X14 duct work, at about 24 feet, airflow begins to drag.

I have had good luck with booster fans in a couple very long runs of ductwork.
For whatever it is worth, I used Cloudline S series fans from AC Infinity. They have been in use for years and work. I had problems with 1 of the fans I installed, it was replaced for free with a newer model by the manufacturer.
